Effect of corosolic acid on postchallenge plasma glucose levels.
Corosolic acid (10 mg/kg) reduced the blood glucose (p<0.05) of KK-Ay mice 4 h after single oral administration when compared with the control group. However, corosolic acid did not change the plasma insulin. The muscle facilitative glucose transporter isoform 4 (GLUT4) translocation from low-density microsomal membrane to plasma membrane was significantly increased in the orally
Corosolic acid significantly inhibited cell viability in both a dose- and a time-dependent manner. Corosolic acid treatment induced S cell-cycle arrest and caused apoptotic death in HeLa cells. We found that Corosolic acid increased in Bax/Bcl-2 ratios by up-regulating Bax expression, disrupted mitochondrial membrane potential and triggered the release of cytochrome c from mitochondria into the cytoplasm. Moreover, Corosolic acid treatment triggered the activation of caspase-8, -9 and -3 in HeLa cells. All these results indicate that Corosolic acid-induced apoptosis is associated with the activation of caspases via a mitochondrial pathway. Taken together, we believe that Corosolic acid could have strong potentials for clinical application in treating human cervix adenocarcinoma and improving cancer Chemotherapy. |
