Use: Diosgenin is the precursor for the semisynthesis of progesterone which in turn was used in early combined oral contraceptive pills. The unmodified steroid has estrogenic activity and can reduce the level of serum cholesterol. Diosgenin also causes an inhibition of the growth of fibroblast-like synoviocytes from human rheumatoid arthritis, with apoptosis induction associated with cyclooxygenase-2 (COX-2) up-regulation. Celecoxib, a selective COX-2 inhibitor, provoked a large decrease in diosgenin-induced apoptosis even in the presence of exogenous prostaglandin E2, whereas interleukin-1β, a COX-2 inducer, strongly increased diosgenin-induced apoptosis of these synoviocytes. Diosgenin inhibited migration and invasion of PC-3 cells by reducing MMPs expression. It also inhibited ERK, JNK and PI3K/Akt signaling pathways as well as NF-?B activity. These findings reveal new therapeutic potential for diosgenin in anti-metastatic therapy. Diosgenin has an obvious antitumor activity on S-180, HepA, U14 transplant mice in vivo and L929, HeLa, MCF cells in vitro.

Notes:
a spirostan found in dioscorea and other plants. the 25s isomer is called yamogenin. solasodine is a natural derivative formed by replacing the spiro-ring with a nitrogen, which can rearrange to solanine.
